Uji Aktivitas Hepatoprotektor Fraksi Etil Asetat Kenikir (Cosmos caudatus) Terhadap Tikus Yang Diinduksi Paracetamol (Hepatoprotector Activity Ethyl Acetate Fraction Of Kenikir (Cosmos caudatus) On Rat Induced By Paracetamol)
Abstract
Abstract: Liver plays a vital role in metabolism and detoxification. Paracetamol in high dose can make acute liver failure (ALF) indicate byliver necrosisand elevated of marker enzyme such us SGOT&SGPT (aminotransferase). Ethanolic extract of kenikir can prevent liver injury caused by paracetamol. Kenikir has good antioxidant activity. Kenikir on ethanolic extract hasIC50 19,43±0,317μg/mL, however ethyl acetate fraction of this has IC50 14,229 μg/mL. With the better antioxidant activity on the ethyl acetate fraction, it give deep opportunity to develop hepatoprotector. This research to know the effective of etyl acetate fraction of kenikir on the rat induced by paracetamol spesificly on aminotrasferase. Hepatotoxicity was induced by paracetamol (2,5 g/kg bw) and the biochemical parameters such as serum glutamate oxalate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT) were studied along with curcuminoid 100 mg/kg BB and etyl acetate fraction of kenikir 281,25; 562,5; 1125 mg/kg bw. All treatments give for seven days.At 7th day, 30 minute after last dose, all rats give paracetamol. At the 9th day, biochemical parameters measured.Etyl acetate fraction of kenikir has hepatoprotector on rat induced by paracetamol. Etyl acetate fraction of kenikir at 1125 mg/kg BB give an optimum effect of hepatoprotective indicate by decrease of SGPT and SGOT (p<0,05)

Abstrak: Liver memainkan peranan yang vital dalam metabolisme dan detoksifikasi senyawa. Penggunaan parasetamol dalam dosis toksik dapat memicu terjadinya kerusakan liver yang ditandai dengan terjadinya nekrosis dan kenaikan kadar enzim seperti SGPT dan SGOT (aminotransferase). Penggunaan ekstrak etanol kenikir terbukti mampu mencegah kerusakan liver yang disebabkan oleh paracetamol.Kenikir terbukti memiliki aktivitas antioksidan yang baik.Kenikir dalam bentuk ekstrak etanol memiliki IC50 19,43±0,317μg/mL, sedangkan fraksi etil asetat memiliki IC50 14,229 μg/mL.Dengan tingginya aktivitas antiosidan kenikir dalam bentuk fraksi etil asetat ini, memberikan peluang yang lebih dalam pengembangkan hepatoprotektor.Penelitian ini dilakukan untuk mengetahui efektivitas fraksi etil asetat kenikir pada tikus yang diinduksi parasetamol. Kerusakan liver diinduksi dengan menggunakan paracetamol (2,5 g/kg BB) dan parameter biokimia yang diteliti meliputi serum glutamate oxalate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT) dengan menggunakan kurkuminoid 100 mg/kg BB) dan fraksi etil asetat kenikir dosis 281,25; 562,5; 1125 mg/kg BB. Semua perlakuan diberikanselama tujuh hari. Pada hari ke-7, 30 menit setelah pemberian dosis terakhir dilanjutkan dengan pemberian paracetamol.Pada hari ke-9, dilakukan pengukuran parameter biokimia.Fraksi etil asetat kenikir mampu memberikan efek hepatoprotektor pada tikus yang diinduksi paracetamol. Fraksi etil asetat kenikir dengan dosis 1125 mg/kg BB menunjukkan efektifitas hepatoprotektor yang signifikan (p<0.05) yang ditunjukkan dengan penuruan parameter biokimia SGPT dan SGOT.
